Objective:
To publish a book on "Analogue-based Drug Discovery".
It should serve as a reference book for medicinal chemists, and afford an easily usable overview on drugs, drug classification and analogue types.

Description:
The book will illustrate the considerable success that has been achieved in the development of new drugs based on the leads provided by established drugs. Such new drugs are often considered deprecatingly as "me-too" products. However, the book will demonstrate that it is part of the very important process whereby drug action is optimized for a given therapeutic effect.

J. Fischer and A. Gere have published the journal article: Pharmazie, 56, 675-682 (2001) on "Timing of analogue research in medicinal chemistry" in order to initiate a book consisting of the following chapters:

1. Drug-Likeness
One of the task group member (J.P.) has recently published an article on "Drugs, Leads, and Drug-Likeness" in Bioorganic and Medicinal Chemistry Letters12, 1647-1650 (2002), which will be developed into a chapter of the book.

2. Analogue Types in Medicinal Chemistry
The Task Group Chairman (J.F.) will continue the above journal article in order to obtain an actual and a more comprehensive overview as a chapter in the book.In this part of the book some examples of successful analogue discoveries will be described by some of the task group members.

3. Drug Classification
J.F. and his present coworkers describe the most important drugs (around 1 000) in tables classified according to their chemical analogy and biological action. A group contains chemical analogues with the same therapeutic area and mechanism of action. The drugs will be ordered according to their patent priority inside a group. The tables will offer important informations on the drugs (main therapeutic areas, mechanism of action, CAS, MW, basic patent, launch, market value).
